MICA is located within the human leukocyte antigen (HLA/MHC) locus, and its exon5 coding transmembrane (TM) region contains a polymorphic microsatellite, classified according to the number of alanine-encoding GCT repeats as A4, A5, A5.1, A6 and A9. A single nucleotide polymorphism (SNP) at codon 129 in exon 3 of the MICA gene, Val129Met or V129M (rs1051792), has been shown to modulate the affinity of MICA to the NKG2D receptor.
