The GAB2 gene, encoding the GRB-associated binding protein 2, has been studied for it's additive effect on the risk of developing late-onset Alzheimer's disease in conjunction with carriers of ApoE4 alleles. [PMID 17553421]

A summary of the increased odds (OR) associated with each SNP is shown below, indicating the risk allele; note that these odds are only relevant to individuals who are (also) ApoE4 carriers, since the research cited did not observe any increased risk for ApoE4 non-carriers:

Note also that the increased risk for developing Alzheimer's seen in this study did not include risk for earlier age at onset.

Overall, an odds ratio of 24.6 for overall genetic risk of developing late-onset Alzheimer's is obtained when both ApoE4 and the GAB2 rs2373115 alleles are present.
